Long-term monitoring datasets are fundamental to understand physical and ecological responses to environmental changes, supporting management and conservation. The data should be reliable, with the sources of bias identified and quantified. CETUS Project is a cetacean monitoring programme in the Eastern North Atlantic, based on visual methods of data collection. This study aims to assess data quality and bias in the CETUS dataset, by 1) applying validation methods, through photographic confirmation of species identification; 2) creating data quality criteria to evaluate the observer's experience; and 3) assessing bias to the number of sightings collected and to the success in species identification. Through photographic validation, the species identification of 10 sightings was corrected and a new species was added to the CETUS dataset. The number of sightings collected was biased by external factors, mostly by sampling effort but also by weather conditions. Ultimately, results highlight the importance of identifying and quantifying data bias, while also yielding guidelines for data collection and processing, relevant for species monitoring programmes based on visual methods.

In the Mediterranean Sea, brown macroalgae represent the dominant species in intertidal and subtidal habitats. Despite conservation efforts, these canopy-forming species showed a dramatic decline, highlighting the urge for active intervention to regenerate self-sustaining populations. For this reason, the restoration of macroalgae forests through transplantation has been recognized as a promising approach. However, the potential stress caused by the handling of thalli has never been assessed. Here, we used a manipulative approach to assess the transplant-induced stress in the Mediterranean Ericaria amentacea, through the analysis of biochemical proxies, i.e., phenolic compounds, lipids, and fatty acids in both transplanted and natural macroalgae over time. The results showed that seasonal environmental variability had an important effect on the biochemical composition of macroalgae, suggesting the occurrence of acclimation responses to summer increased temperature and light irradiance. Transplant-induced stress appears to have only amplified the biochemical response, probably due to increased sensitivity of the macroalgae already subjected to mechanical and osmotic stress (e.g., handling, wounding, desiccation). The ability of E. amentacea to cope with both environmental and transplant-induced stress highlights the high plasticity of the species studied, as well as the suitability of transplantation of adult thalli to restore E. amentacea beds.

Cycling of organic carbon in the ocean has the potential to mitigate or exacerbate global climate change, but major questions remain about the environmental controls on organic carbon flux in the coastal zone. Here, we used a field experiment distributed across 28° of latitude, and the entire range of 2 dominant kelp species in the northern hemisphere, to measure decomposition rates of kelp detritus on the seafloor in relation to local environmental factors. Detritus decomposition in both species were strongly related to ocean temperature and initial carbon content, with higher rates of biomass loss at lower latitudes with warmer temperatures. Our experiment showed slow overall decomposition and turnover of kelp detritus and modeling of coastal residence times at our study sites revealed that a significant portion of this production can remain intact long enough to reach deep marine sinks. The results suggest that decomposition of these kelp species could accelerate with ocean warming and that low-latitude kelp forests could experience the greatest increase in remineralization with a 9% to 42% reduced potential for transport to long-term ocean sinks under short-term (RCP4.5) and long-term (RCP8.5) warming scenarios. However, slow decomposition at high latitudes, where kelp abundance is predicted to expand, indicates potential for increasing kelp-carbon sinks in cooler (northern) regions. Our findings reveal an important latitudinal gradient in coastal ecosystem function that provides an improved capacity to predict the implications of ocean warming on carbon cycling. Broad-scale patterns in organic carbon decomposition revealed here can be used to identify hotspots of carbon sequestration potential and resolve relationships between carbon cycling processes and ocean climate at a global scale.

Accurate species identification often relies on public repositories to compare the barcode sequences of the investigated individual(s) with taxonomically assigned sequences. However, the accuracy of identifications in public repositories is often questionable, and the names originally given are rarely updated. For instance, species of the Sea Lettuce (Ulva spp.; Ulvophyceae, Ulvales, Ulvaceae) are frequently misidentified in public repositories, including herbaria and gene banks, making species identification based on traditional barcoding unreliable. We DNA barcoded 295 individual distromatic foliose strains of Ulva from the North-East Atlantic for three loci (rbcL, tufA, ITS1). Seven distinct species were found, and we compared our results with all worldwide Ulva spp. sequences present in the NCBI database for the three barcodes rbcL, tufA and the ITS1. Our results demonstrate a large degree of species misidentification, where we estimate that 24%-32% of the entries pertaining to foliose species are misannotated and provide an exhaustive list of NCBI sequences reannotations. An analysis of the global distribution of registered samples from foliose species also indicates possible geographical isolation for some species, and the absence of U. lactuca from Northern Europe. We extended our analytical framework to three other genera, Fucus, Porphyra and Pyropia and also identified erroneously labelled accessions and possibly new synonymies, albeit less than for Ulva spp. Altogether, exhaustive taxonomic clarification by aggregation of a library of barcode sequences highlights misannotations and delivers an improved representation of species diversity and distribution.

During high tide, macroalgae are submersed, facing adequate environmental conditions, however, at low tide, these species can be exposed to high UV radiation and desiccation, leading to an overproduction of reactive oxygen species, causing oxidative stress. Since intertidal organisms present differential sensitivity to abiotic fluctuations, this study aimed to evaluate the physiological responses [photosynthetic pigments, hydrogen peroxide (H2O2), lipid peroxidation (LP), and thiols and proline] of three macroalgae, from different intertidal levels, towards tidal regimes. Samples of Pelvetia canaliculata, Ascophyllum nodosum, and Fucus serratus were collected from beaches located on the southern limit of distribution in periods of potential stress (Summer and Spring), under low and high tide. The photosynthetic pigments of P. canaliculata and F. serratus were generally higher during low tide, and the oxidative damage evidenced by H2O2 and LP increased in the Summer, while A. nodosum showed greater oxidative damage in the Spring. While thiol content did not change, proline levels were species- and tidal-specific among sampling dates. P. canaliculata presented higher resilience to unfavorable conditions, while F. serratus was the most sensitive species. The physiological responses analyzed were species-specific, pointing to the high susceptibility of low intertidal organisms to expected extreme climatic events.

Vestigial organs are historical echoes of past phenotypes. Determining whether a specific organ constitutes a functional or vestigial structure can be a challenging task, given that distinct levels of atrophy may arise between and within lineages. The mammalian pineal gland, an endocrine organ involved in melatonin biorhythmicity, represents a classic example, often yielding contradicting anatomical observations. In Xenarthra (sloths, anteaters, and armadillos), a peculiar mammalian order, the presence of a distinct pineal organ was clearly observed in some species (i.e., Linnaeus's two-toed sloth), but undetected in other closely related species (i.e., brown-throated sloth). In the nine-banded armadillo, contradicting evidence supports either functional or vestigial scenarios. Thus, to untangle the physiological status of the pineal gland in Xenarthra, we used a genomic approach to investigate the evolution of the gene hub responsible for melatonin synthesis and signaling. We show that both synthesis and signaling compartments are eroded and were probably lost independently among Xenarthra orders. Additionally, by expanding our analysis to 157 mammal genomes, we offer a comprehensive view showing that species with very distinctive habitats and lifestyles have convergently evolved a similar phenotype: Cetacea, Pholidota, Dermoptera, Sirenia, and Xenarthra. Our findings suggest that the recurrent inactivation of melatonin genes correlates with pineal atrophy and endorses the use of genomic analyses to ascertain the physiological status of suspected vestigial structures.

The ancestors of Cetacea underwent profound morpho-physiological alterations. By displaying an exclusive aquatic existence, cetaceans evolved unique patterns of locomotor activity, vigilant behaviour, thermoregulation and circadian rhythmicity. Deciphering the molecular landscape governing many of these adaptations is key to understand the evolution of phenotypes. Here, we investigate Cortistatin (CORT), a neuropeptide displaying an important role in mammalian biorhythm regulation. This neuropeptide is a known neuroendocrine factor, stimulating slow-wave sleep, but also involved in the regulation of energy metabolism and hypomotility inducement. We assessed the functional status of CORT in 359 mammalian genomes (25 orders), including 30 species of Cetacea. Our findings indicate that cetaceans and other mammals with atypical biorhythms, thermal constraints and/or energy metabolism, have accumulated deleterious mutations in CORT. In light of the pleiotropic action of this neuropeptide, we suggest that this inactivation contributed to a plethora of phenotypic adjustments to accommodate adaptive solutions to specific ecological niches.

The CETUS project is a cetacean monitoring program that takes advantage of cargo ships to undertake survey routes between Continental Portugal, Macaronesian archipelagos and West Africa. From 2012 to 2017, over 50 volunteers participated in the program, actively surveying more than 124.000 km, mostly beyond national jurisdictions in the high seas, for which little or no previous data existed. In total, the collection comprises 3058 georeferenced transect lines and 8913 positions, which are associated with 2833 cetacean sightings, 362 occurrences of other pelagic megafauna, 5260 estimates of marine traffic and 8887 weather observations. This dataset may provide new insights into the distribution of marine mammals in the Eastern North Atlantic and was published following the OBIS-ENV-DATA format (with the most recent biodiversity data standards at the time of writing). Consequently, it may serve as a model for similar visual line transect data collections yet to be published.

Neurodegenerative processes involve numerous and closely related events that ultimately culminate in neuronal cell injury. The aim of this study was (i) to assess, for the first time, the neuroprotective potential of acetone extracts of six edible species of Ochrophyta, by evaluating their cholinesterase and lipoxygenase inhibitory activity in cell-free assays, as well as their capacity to attenuate glutamate-induced toxicity in neuronal (SH-SY5Y) cells, and (ii) to try to relate the chemical composition of the extracts with their biological activity, evaluating also the effect of the main compounds thereof. In spite of a modest cholinesterase inhibition, a dose-dependent response towards lipoxygenase was found for all macroalgae extracts. At non-cytotoxic concentrations, the extracts from Fucus serratus Linnaeus and Saccharina latissima (Linnaeus) C.E. Lane, C. Mayes, Druehl & G.W. Saunders were able to improve the viability of glutamate-insulted SH-SY5Y cells. These results encourage further studies for a more detailed understanding of the mechanisms beyond the documented biological activities, and point to the potential interest of the selected seaweed species and their extracts as promising candidates for in vivo studies.

The aims of this study were to explore mollusc assemblages associated with the non-indigenous macroalga Asparagopsis armata, to compare them with those on other macroalgae at the study region and to explore potential differences on mollusc assemblages between two regions in the Atlantic coast of the Iberian Peninsula, where A. armata is present. To achieve this, at each region, four intertidal shores were sampled. Twenty-nine mollusc species were reported and thus, A. armata harboured similar or higher diversity than other annual macroalgae in this area. When compared with perennial macroalgae, results depend on the species and studied area. Moreover, significant differences in structure of mollusc assemblages between the two studied regions were found. However, these were due to differences in the relative abundance of species rather than the presence of exclusive species at each region.

We establish baseline knowledge of abundance, diversity and multivariate structure of macrobenthos from shallow sublitoral soft bottoms in the North Portuguese coast and elucidate main environmental factors that shape their spatial patterns. In this area distribution of soft bottoms is patchy, surrounded by boulders and rocky substrates. This particular landscape and the lack of significant anthropogenic disturbances are values for the conservation of this habitat. Sediment and physicochemical properties of the water column were studied to provide models for each studied macrobenthic variable. Our models highlighted that most of variation (59%-72%) in macrobenthic spatial patterns was explained by the studied environmental variables. Sedimentary variables were more relevant that those of the water column. Therefore, disturbances affecting sedimentary environment could cause dramatic changes in macrobenthic assemblages because of the limited availability of soft bottoms in the area. In this way, results are useful to adopt right management and conservation strategies.

Kelp forests (Order Laminariales) form key biogenic habitats in coastal regions of temperate and Arctic seas worldwide, providing ecosystem services valued in the range of billions of dollars annually. Although local evidence suggests that kelp forests are increasingly threatened by a variety of stressors, no comprehensive global analysis of change in kelp abundances currently exists. Here, we build and analyze a global database of kelp time series spanning the past half-century to assess regional and global trends in kelp abundances. We detected a high degree of geographic variation in trends, with regional variability in the direction and magnitude of change far exceeding a small global average decline (instantaneous rate of change = -0.018 y-1). Our analysis identified declines in 38% of ecoregions for which there are data (-0.015 to -0.18 y-1), increases in 27% of ecoregions (0.015 to 0.11 y-1), and no detectable change in 35% of ecoregions. These spatially variable trajectories reflected regional differences in the drivers of change, uncertainty in some regions owing to poor spatial and temporal data coverage, and the dynamic nature of kelp populations. We conclude that although global drivers could be affecting kelp forests at multiple scales, local stressors and regional variation in the effects of these drivers dominate kelp dynamics, in contrast to many other marine and terrestrial foundation species.

Spatial variability of environmental factors and macrobenthos, using species and functional groups, was examined over the same scales (100s of cm to >100 km) in intertidal sediments of two transitional water systems. The objectives were to test if functional groups were a good species surrogate and explore the relationship between environmental variables and macrobenthos. Environmental variables, diversity and the multivariate assemblage structure showed the highest variability at the scale of 10s of km. However, abundance was more variable at 10s of m. Consistent patterns were achieved using species and functional groups therefore, these may be a good species surrogate. Total carbon, salinity and silt/clay were the strongest correlated with macrobenthic assemblages. Results are valuable for design and interpretation of future monitoring programs including detection of anthropogenic disturbances in transitional systems and propose improvements in environmental variable sampling to refine the assessment of their relationship with biological data across spatial scales.

The biomass yield potential of Mastocarpus stellatus, a commercially attractive carrageenophyte for foods and pharmaceutics, was investigated by cultivating the seaweeds in the nutrient-rich outflow of a commercial fish farm. Results from two consecutive 4 weeks experiments indicate that the cultivation of this seaweed produces a mean biomass of 21 to 40.6 gDW m(-2) day(-1) depending on the time of the experiment. DRIFT and CP-MAS NMR analyses of seaweeds indicate that cultivation during May affected quantitatively the seaweeds chemistry, and thus the chemical and gelling properties of native extracts of kappa/iota-hybrid carrageenan (KI). Overall, algal growth leads to the production of more sulphated KI, the percentage increase varying between 27% and 44% for the two experiments. However, alkali treatment of seaweeds before extraction reduces the variations in gelling properties of KI induced by the algal growth. This study demonstrates the capacity of growing M. stellatus in an integrated multi-trophic aquaculture system for the sustainable production of high value polysaccharides.

The allocation of resources to different life-history traits should represent the best compromise in fitness investment for organisms in their local environment. When resources are limiting, the investment in a specific trait must carry a cost that is expressed in trade-offs with other traits. In this study, the relative investment in the fitness-related traits, growth, reproduction and defence were compared at central and range-edge locations, using the seaweed Ascophyllum nodosum as a model system. Individual growth rates were similar at both sites, whereas edge populations showed a higher relative investment in reproduction (demonstrated by a higher reproductive allocation and extended reproductive periods) when compared to central populations that invested more in defence. These results show the capability of A. nodosum to differentially allocate resources for different traits under different habitat conditions, suggesting that reproduction and defence have different fitness values under the specific living conditions experienced at edge and central locations. However, ongoing climate change may threaten edge populations by increasing the selective pressure on specific traits, forcing these populations to lower the investment in other traits that are also potentially important for population fitness.

Habitat structure is a primary factor determining the organism distribution. Here, two native and one invasive macroalgal species, apparently different in morphology, were sampled to examine the effects of habitat complexity on the abundance (N), taxon richness (S) and structure of their associated epifaunal assemblages by means of univariate and multivariate techniques. Dry weight and fractal measures were used as proxies of habitat quantity and habitat architecture respectively. Results revealed significant differences in the complexity and in N, S and the structure of epifaunal assemblages among macroalgae and significant correlations between complexity and epifauna. Results suggested that, beside the effect of habitat quantity, the habitat architecture also seems to play a significant role in shaping epifaunal assemblages. Complexity of the studied invasive macroalga significantly differed from that of native species and hosted also different assemblages. Therefore, our findings suggest that invasive macroalgae, if structurally different from native species, induce changes in the associated epifauna.

We investigated effects of subtle nutrient enrichment and metal pollution on different levels of biological organization (i.e. whole assemblage, population and individual) of species-rich assemblages. We used rockpools as model system, applying a multi-factorial sampling design to test hypotheses on differences between disturbed and reference locations. Results indicated that disturbed and reference locations supported similar assemblages, as well as individual fitness-related life-traits were ineffective to discriminate between the two conditions. In contrast, assemblages responded to pollution through a reduction of the abundance of sensitive species and a proliferation of tolerant species, although these alterations were detectable only once the influence of dominant taxa was down-weighed by data transformation. Present findings suggest that, contrarily to individual level variables, assemblage structure after data transformation and patterns of distribution and abundance of differently sensitive taxa would be a powerful tool to detect effects of subtle pollution on species-rich assemblages.

Metals (Cd, Cr, Cu, Fe, Mn and Zn) in coastal seawaters and soft tissues of macroalga Fucus spiralis from the northwest coast of Portugal were determined to assess spatial variations of metal bioavailabilities and bioaccumulation factors to compare different ecological quality classifications. Both coastal seawaters and soft tissues of F. spiralis showed significant spatial variations in their metal concentrations along the coast. The macroalgae F. spiralis accumulated more efficiently Cd, Mn and Zn and showed low bioaccumulation factors to Cr, Cu and Fe. Regarding the metal guidelines of the Norwegian Pollution Control Authority, the entire northwest (NW) coast of Portugal in April 2013 should be classified as 'class I--unpolluted' for all metals, except in Ave for Cu ('class II--moderately polluted') and Cavado for Cd and Cu ('class II-moderately polluted'), revealing the low metal bioavailabilities of these seawaters. As there were always significant positive correlations between all metals in seawaters and F. spiralis, this macroalga species was considered a suitable monitoring tool of metal contamination in the NW coast of Portugal and a useful aquatic organism to be included in the European Environmental Specimen Banks in order to establish a real-time environmental monitoring network under the European Water Framework Directives.

Persistence of populations at range edges relies on local population dynamics and fitness, in the case of geographically isolated populations of species with low dispersal potential. Focusing on spatial variations in demography helps to predict the long-term capability for persistence of populations across the geographical range of species' distribution. The demography of two ecological and phylogenetically close macroalgal species with different life history characteristics was investigated by using stochastic, stage-based matrix models. Populations of Ascophyllum nodosum and Fucus serratus were sampled for up to 4 years at central locations in France and at their southern range limits in Portugal. The stochastic population growth rate (λ(s)) of A. nodosum was lower and more variable in central than in southern sites whilst for F. serratus this trend was reversed with λ(s) much lower and more variable in southern than in central populations. Individuals were larger in central than in southern populations for both species, which was reflected in the lower transition probabilities of individuals to larger size classes and higher probability of shrinkage in the southern populations. In both central and southern populations elasticity analysis (proportional sensitivity) of population growth rate showed that fertility elements had a small contribution to λ(s) that was more sensitive to changes in matrix transitions corresponding to survival. The highest elasticities were found for loop transitions in A. nodosum and for growth to larger size classes in F. serratus. Sensitivity analysis showed high selective pressure on individual growth for both species at both locations. The results of this study highlight the deterministic role of species-specific life-history traits in population demography across the geographical range of species. Additionally, this study demonstrates that individuals' life-transitions differ in vulnerability to environmental variability and shows the importance of vegetative compared to reproductive stages for the long-term persistence of populations.

Native agars from Gracilaria vermiculophylla produced in sustainable aquaculture systems (IMTA) were extracted under conventional (TWE) and microwave (MAE) heating. The optimal extracts from both processes were compared in terms of their properties. The agars' structure was further investigated through Fourier transform infrared and NMR spectroscopy. Both samples showed a regular structure with an identical backbone, ß-d-galactose (G) and 3,6-anhydro-α-l-galactose (LA) units; a considerable degree of methylation was found at C6 of the G units and, to a lesser extent, at C2 of the LA residues. The methylation degree in the G units was lower for MAE(opt) agar; the sulfate content was also reduced. MAE led to higher agar recoveries with drastic extraction time and solvent volume reductions. Two times lower values of [η] and M(v) obtained for the MAE(opt) sample indicate substantial depolymerization of the polysaccharide backbone; this was reflected in its gelling properties; yet it was clearly appropriate for commercial application in soft-texture food products.
